IML3: part of the CCAN complex
CCAN is a multi-protein complex. [5] It acts as a scaffold in kinetochore formation [6] by changing conformation during specific stages of the cell cycle to recruit microtubules onto the chromatids. [7]
In S. cerevisiae, the CCAN protein IML3 is homologous to vertebrate CENP-L, and interacts with CHL4 (CENP-N) and CTF3 (CENP-I). [2]
IML3 connects the central and outer kinetochore [1,8] and mediates the construction of metaphasic chromosomes by binding to CHL4. [9]
In 2002, Cheeseman et al. purified the kinetochore proteins through altered tandem affinity purification. This is suitable for isolating protein complexes instead of individual proteins. After mass spectrometry analysis, the CHL4/IML3 subcomplex was identified as a part of the CCAN complex. [10]
